Your question is too broad for an answer shorter than a textbook. I would recommend obtaining the AIAG MSA Manual and Donald Wheeler's book Evaluating the Measurement Process III Using Imperfect Data.

Then when you have more specific questions, we would be glad to help you.

There are also many MSA files in this forum that you can search and download. However, you do need to verify that the file has been updated to the latest calculations in the MSA Manual. There is a Ford Appoved file in the forum with validation data and the expected calculation results that you can use to validate your spreadsheet or software.
