I’m just curious.. What is its significance to U.S.? What symbolizes his costume? What does the pointing right finger mean?

Uncle Sam is actually based on a real person Sam Wilson. Sam Wilson was a guy that provided the US army with beef barrels in the war of 1812. On the barrels was marked U.S. for United States, but the soldiers adopted the acronym as "Uncle Sam".

The character portrayed in the posters doesnt actually look like Sam Wilson, but was adopted based on that history i just explained by the US government, during WW1 for recruitment purposes.

The cartoon was the idea of Thomas Nast, and I guess drawn to look like the personification of American patriotism... in my opinion he looks like Abe Lincoln, who it could of been modeled after. Because Abe Lincoln is often heralded as the greatest American ever...
