I think there is no written response I could give you that's better than a reference to a good book. One of the best looking, well-rounded and practical information sources I've seen is in David Mann's book Creating a Lean Culture. It addresses the "four winds" that I like to list as man, material, machine and method. It even goes into the very human problems of dealing with resistance and low performers. No, I'm not affiliated though I do carry on like a cheerleader. Note: I have the 2005 edition, which is available used for about $10. I haven't seen the 2nd edition (2010).

You could consider this a Lean Six Sigma project ... its great !

We have plenty of threads on the said subject. One of them is a recently posted response now - What is the difference between six sigma and Lean six sigma ?

Without knowing your business it is not easy to give a good answer but whatever you introduce or do involve as many people as possible and do not try something anything too complicated.

I would also recommend promoting the benefits gained internally to ensure the initiative gains momentum.

We have just held a workshop to re-layout our manufacturing area, involving all personnel working in there with great success. This enable us to improve flow through the facility and introduce FOD Compliance to NAS412 at the same time.

From this we have benefited in reduction of labour times and because the staff were all involved they have really got behind the process and embraced what we were trying to achieve

In addition to what others have suggested, also try implementing ISO 9004: 2009 in a systematic manner for sustained success of the organization.
