Not being your customer, and having done only a small amount of work with aerospace suppliers I don't know for sure. I could see it as:
- a manual (e.g., set of procedures, forms, etc.) that spells out the processes used for developing and validating designs (and design changes), basically equivalent to section 7.3 in ISO 9001 (or AS 9100).
- a set of guidelines used for designing specific products (e.g., formulas to use, minimum &/or maximum limits of capability, test methods).
Best thing to do would be to ask your customer what they mean by it.

My guess is that what's meant is a controlled compilation of design-related policies and practices.
Have you asked your engineers how they learned your organization's work methods and documentation practices when they were hired, and what they use as a work-method reference when they're assigned a task that they haven't done before? Perhaps such a manual, or list of individual documents, already exists external to your QS.
My guess would be that if such policies and practices don't exist now, creating a document that purports to present such information as your organization's design practice would be less than fully factual.
