Freeze-Dried Powder Sterility Test using the Membrane Filtration Technique

After long time we are manufacture again a batch of freeze-dired powder, but when perfom sterility test using the membrane filtration technique, the membrane is saturated and does not allow the passage of the liquid and the water determination test can not be defined. I suppose has large quantity of water so it is involve the stability of the product.
Some part of the proccess manufature is involved?
Can I change themembrane filtration technique by direct inoculation?
